چکیده

Exosomes are small vesicles released by various cell types, including tumor cells, as a result of fusion of the multivesicular bodies with the plasma membrane. Exosomes can interact with target cells and mediate different processes, such as, tumor progression and intercellular transfer of proteins and RNA. Although heavily glycosylated forms of some proteins were found in exosomes, the role of glycosylation on protein sorting to exosomes and on the recognition and uptake of these vesicles by cells is still not known. The analysis of the glycosylation profiles of cellular extracts and exosomes from ovarian carcinoma SKOV3 cells, using lectins with different specificities, showed higher levels of protein glycosylation, in particular, sialylation in exosomes. SKOV3 cells were found to internalize SKOV3 derived exosomes, labeled with carboxyfluoresceine diacetate succinimidyl-ester (Exos-CFSE), through an energydependent process that was almost completely inhibited at 4 oC. Digestion of Exos-CFSE and SKOV3 cells with proteinase K resulted in considerably lower levels of uptake indicating that proteins were required, at least in part, for internalization. Exos-CFSE uptake was slightly increased after exosomes desialylation with neuraminidase and blocking of cell surface glycan-receptors with different monosaccharides had no effect when compared with the control D-glucose. In conclusion, the increase in protein glycosylation was not essential for exosomes recognition and uptake by target cells. The presence of some heavily glycosylated proteins in exosomes might be a consequence of their enrichment in specific domains of the plasma membrane from where exosomes emerge. The effect of exosomes derived from human ovarian epithelial cancer cells on the secretion of AMH and Inhibin in granulosa cells

Exosomes are secreted by different types of cells and known as biological packages. Exosomes have significant role in intercellular communications and involved in the development and progression of various diseases such as cancer. Inhibin B and anti-mullerian hormone (AMH) are markers of granulosa cell tumors (GCT) and due to the role of exosomes in the progression of cancer, in this experiment...

Assessment of Antitumor Activity of Vinca herbacea on Human Ovarian Cancer Cell Line

Background: It seems that Vinca. herbacea has an anti-tumor effect. Here, the immunotherapeutic effect of this compound is assessed against human ovarian cancer (SKOV3) cells because of the high incidence of this tumor in women. Materials and Methods: The cytotoxic activity of V. herbacea extract against human ovarian cancer (SKOV3) cells was determined by MTT assay. The apoptosis-inducing pote...

Downregulation of miR-17~92 Expression Increase Paclitaxel Sensitivity in Human Ovarian Carcinoma SKOV3-TR30 Cells via BIM Instead of PTEN

To better understand the molecular mechanisms of paclitaxel resistance in ovarian carcinoma, we evaluated the expression of miRNAs using miRNA microarray between human ovarian carcinoma SKOV3 cells and paclitaxel resistant SKOV3-TR30 cells. Results showed that 69 miRNAs were upregulated while 102 miRNAs were downregulated in SKOV3-TR30 cells. Using real-time PCR, we further clarified that miR...

Exosomes in the ascites of ovarian cancer patients: origin and effects on anti-tumor immunity.

This study was performed to identify the origin of the ascites-derived exosomes from patients with ovarian cancer and to observe the effect of exosomes on anti-tumor immunity. Exosomes were isolated from the ascites of patients with ovarian epithelial cancer by ultracentrifugation plus density gradient centrifugation. The origin of exosomes was identified by immunoelectronmicroscopy (IEM). The ...

Emblica officinalis extract downregulates pro-angiogenic molecules via upregulation of cellular and exosomal miR-375 in human ovarian cancer cells

Ovarian cancer (OC) is highly resistant to current treatment strategies based on a combination of surgery, chemotherapy and radiation therapy. We have recently demonstrated the anti-neoplastic effect of Amla extract (Emblica officinalis, AE) on OC cells in vitro and in vivo. We hypothesized that AE attenuates growth of OC through microRNA (miR)-regulated mechanism(s). The inhibitory effect of A...
